Inside the World's First Age Reversal Trial

In this episode of Lifespan, Dr. David Sinclair, A.O., Ph.D. explores the science of eye aging, vision loss, and emerging strategies to preserve vision throughout life. Dr. Sinclair shares an exclusive update on ER-100, including his team's successful restoration of vision in non-human primates and the launch of the world's first FDA-cleared age reversal human clinical trial. This Phase 1 clinical trial will evaluate the safety of epigenetic cellular restoration as a therapy. Additionally, drawing on decades of research, Dr. Sinclair explains why the eyes may offer one of the earliest windows into biological aging, how everyday factors such as sleep position, alcohol consumption, and intraocular pressure influence long-term eye health, and what the latest evidence reveals about nutrition, supplements, and the connection between the eyes and the brain. Whether you're interested in longevity, brain health, or protecting your eyesight, this episode provides an evidence-based look at the biology of vision and the future of regenerative medicine.

Fasting as a Path to Longevity: The Facts

In this episode of Lifespan, Dr. David Sinclair, A.O., Ph.D. examines the science of calorie restriction and fasting, two of the most studied interventions for healthy aging. Drawing on decades of research, Dr. Sinclair explains how eating less – and eating less often – affects the biology of aging, why our evolutionary past may predispose many of us to weight gain, and what the latest human and animal studies reveal about lifespan, metabolism, and disease risk. Whether you're interested in longevity, metabolism, or intermittent fasting, this episode provides an evidence-based look at one of the most powerful strategies for supporting health and longevity.

NMN, NR, Resveratrol, Metformin & Other Longevity Molecules | Lifespan with Dr. David Sinclair #4

In this week's episode, Dr. David Sinclair and co-host Matthew LaPlante zero in on drugs and supplements that have been reported to combat aspects of aging. They share the latest experimental and clinical data for NAD boosters (these being NR, NMN, NAD IV drips and shots), resveratrol, fisetin, quercetin, rapamycin, spermidine, metformin, and berberine. Given the abundance of data available, a special focus is placed on the NAD precursors nicotinamide riboside and nicotinamide mononucleotide. Known mechanisms, limitations, and/or side effects associated with these molecules are additionally highlighted.

The Science of Looking Younger, Longer | Lifespan with Dr. David Sinclair #6

In Episode 6 of the Lifespan podcast, Dr. David Sinclair and co-host Matthew LaPlante discuss cosmetic aging and how to improve skin, nails, and hair. They talk about why superficial aging occurs and how external signs of aging are often a reflection of biological age. The latest science behind various beautifying therapies is highlighted, including newer interventions like low-level laser therapy and platelet-rich plasma injections.

Biotracking, Age Reversal & Other Advanced Health Technologies | Lifespan with Dr. David Sinclair #8

In the final episode of this season, Dr. David Sinclair and Matthew LaPlante focus on current and near-future technologies relevant to health and aging. In addition to discussing the utility of wearable sensors and biological age measurements, they highlight innovative research aimed at reversing biological age. The societal effects of therapies that successfully extend healthspan and/or lifespan are also considered.

The Science of Keeping the Brain Healthy | Lifespan with Dr. David Sinclair #7

In this episode of the Lifespan podcast, Dr. David Sinclair and co-host Matthew LaPlante dissect the topic of brain aging. They explore evidence suggesting that the brain ages more slowly than other parts of the body and highlight how cognitive function is impacted by aging. Different interventions aimed at preserving brain health are also discussed, including a plant-based diet, exercise, metformin, NAD boosters, and sufficient sleep.

Exercise, Heat, Cold & Other Stressors for Longevity | Lifespan with Dr. David Sinclair #3

In Episode 3 of the Lifespan Podcast, Dr. David Sinclair and Matthew LaPlante dive deeply into the science of non-dietary interventions that mimic adversity and promote health. They begin by highlighting how different types of physical activity (i.e., low-intensity aerobic exercise, high-intensity aerobic exercise, and weight training) protect against age-related disease and enhance longevity. David and Matthew additionally highlight the latest evidence behind hyperbaric oxygen therapy, cold therapy, and heat therapy. As they discuss different adversity mimetics, they also explain how these interventions influence aging at the molecular and physiological levels.

The Science Behind Why We Age

In this episode, Dr. David Sinclair and co-host Matthew LaPlante discuss why we age. In doing so, they discuss organisms that have extreme longevity, the genes that control aging (mTOR, AMPK, Sirtuins), the role of sirtuin proteins as epigenetic regulators of aging, the process of "ex-differentiation" in which cells begin to lose their identity, and how all of this makes up the "Information Theory of Aging", and the difference between "biological age" and "chronological age" and how we can measure biological age through DNA methylation clocks.

Such hopeful research! My daughter’s optic nerves were crushed by a brain tumor, and listening to you talk about your aging research beginning with fixing loss of sight is just amazing. Brought me to tears thinking of the possibilities for her, hopefully sooner than later. I loved every episode of this series and can’t wait for the next season.

If you are interested in aging, this is a must listen. Even if you’ve read Dr. Sinclair’s book “Lifespan,” a lot of advancements have been made in the brief time since it was written. This podcast offers easy to understand explanations of the science of aging and practical advice...

Best podcast I've heard and I'm a podcast addict. High level information, newest research and things you just won't hear in other places but broken up in a way that I can understand.

I have completely restored my deep sleep cycle by following the suggested intermittent fasting and getting all my eating finished before 2pm. Can’t thank these guys enough, wish I could use more of their wisdom, awaiting season 2 with excitement.

If I could give your podcast 10 Stars, I would! I’ve learned a ridiculous amount of information about my own body and the science of aging. My parents died early (age 65 and 67) from undiagnosed cancer and a sudden aortic aneurysm.
